MicroRNA related anti-tumour pathways analysis in HeLa cell lines dysregulated by lupeol

Abstract: Lupeol attracted lots of research attention because of its anti-tumour activity. This study investigates the complete small RNA sequencing data analysis process, including miRNA annotation, novel miRNA prediction, dysregulated miRNA identification, miRNA target prediction, and target gene ontology analysis, and compares the complete miRNA profile of HeLa cell lines between lupeol treatment group and control group. The experimental results indicate that the predicted miRNAs are highly associated with carcinogenesis. By integrating miRNA profile with transcriptome and proteome, we select out the miRNA-target pairs for further real time quantitative PCR identification. MiR-183-96-182 cluster is significantly down-regulated in HeLa cells treated by lupeol, and KIAA1199 mRNA is the target of miR-183, while PBX1 mRNA is the target of miR-182. We conclude that lupeol participates in various molecular pathways, of which Wnt signalling pathway and BAX verify the previous research results.
