Study on vitro transdermal administration of surface anaesthetics based on dextran-based nano-injection lipid lidocaine

Abstract: The paper explored the in vitro transdermal performance and in vivo characteristics of surface anaesthetics containing lidocaine (LID-HLD-BNs) using dextran-based nanoliposome as a carrier. Methods: The paper used lidocaine-encapsulated traditional liposomes (LID-CLs) and lidocaine hydrochloride injection (LID-IJ) as controls, using isolated mouse abdominal skin, using Franz diffusion cell, high-performance liquid Chromatographic method was used to conduct in vitro transdermal experiments to observe the transdermal performance of LID-HLD-BNs. Using calcian as a probe, using laser confocal microscope (CLSM), with calcian-loading traditional liposomes (calcian-CLs) and free calcian as controls, observe the liposome preparation (calcian-HLD-BNs) Fluorescence penetration in the abdomen of living mice. Results: The study found that LID-HLD-BNS' cumulative transdermal volume per unit area and transdermal rate were higher than LID-CLs and LID-IJ. The transdermal rates of LID-IJ, LID-CLs, and LID-HLD-BNs were 39.15, 79.04, and 142.86 μg/(cm2·h), respectively, and the difference was statistically significant (P < 0.01). In the in vivo fluorescence transdermal experiment, the skin penetration speed and depth of the calcian-HLD-BNs group were significantly greater than those of the calcian-CLs group and the free calcian group. Conclusion: Dextran-based nanoliposomes have good transdermal permeability.
